Posttranslational modifications (PTM) of histones, such as methylation, acetylation, and phosphorylation play a significant role in controlling gene expression and DNA repair. Methylation of lysine has been of particular interest due to its slow turnover in vivo and association with several disease phenotypes, including cancer. The lysine residues can be mono-, di- or tri-methylated. Methylation itself is not sufficient to effect changes in transcriptional activity. Other proteins recognize specific histone methylation marks essentially serving as code readers. These chromatin-associated proteins are gaining attention as potential therapeutic targets for epigenetic related diseases. Biotinylated histone derived peptides have been used to study effectors proteins translating PTMs into functional changes.
