Neuropeptide Y (NPY) is a 36-amino acid widely distributed neuropeptide that mediates physiological effects via the interaction with six different receptors [Y(1)-Y(6)]. NPY plays an important role in the regulation of energy homeostasis. It modulates the sympathetic nervous system to control blood pressure and also plays a role as a physiological stabilizer of neural activity in circuits involved in the regulation of arousal and anxiety. NPY stimulates feeding desire, increases body weight and anabolic hormone levels by activating the NPY-Y(1) and Y(5) receptors in the hypothalamus. It potently stimulates food intake not only by increasing NPY signaling, but by reducing melanocortin signaling. It possesses inhibitory action on the gonadotropic and somatotropic systems and may have an effect on the properties of vascular endothelial cells such as pro-angiogenic effects and potentiation of noradrenaline-induced vasoconstriction. Central administration of NPY has also been shown to decrease sympathetic nervous system activity and to suppress thermogenesis in brown adipose tissue. A high density of NPY receptors in breast and ovarian cancers suggests a role of NPY in tumor biology.
