Peptide Tyrosine Tyrosine (PYY), Neuropeptide Y (NPY) and Pancreatic Polypeptide (PP) are anorectic gut hormones belonging to the PP fold peptide family. PYY shares over 70% sequence identity with NPY and is a 36-amino acid regulatory gut hormone present in endocrine cells of the intestine. Specific PYY receptors have been reported in brain tissue, spleen, vascular smooth muscle, neuroendocrine cell lines, chief cells, and mucosa of the small and large intestine. Full-length PYY binds with high affinity to all Y receptors. PYY acts both as an endocrine and a paracrine agent. It is secreted from intestinal L-cells in response to dietary carbohydrate and fat. The biological action of this peptide includes inhibition of gut motility and gastrointestinal and pancreatic secretions. It also acts as a vasoconstrictor. These effects are thought to be mediated both directly in the periphery and centrally via the area postrema and the nucleus of the solitary tract. The majority of PYY found in the cells and present in the circulation is in the PYY(3-36) form. This Y2R agonist is released from the gastrointestinal tract postprandially in proportion to the calorie content of a meal. Like adipocyte hormone leptin, PYY reduces food intake by modulating appetite circuits in the hypothalamus. PYY deficiency may contribute to the pathogenesis of obesity.
