AnaSpec is pleased to announce the release of two (2) new SensoLyte® assay kits for the measurement of Matriptase and Pin1 activity and 38 Amino Acid Preloaded Hydrazide Resins.

Matriptase (also known as MT-SP1, ST14, TADG-15 and epithin) is a trypsin-like protease from the family of type II transmembrane serine proteases. The spectrum of known Matriptase substrates includes extracellular matrix proteins, cell adhesion molecules, ion channels, growth-factor-like proteins and other proteases. Its actions can result in protein processing, activation or degradation. Matriptase is widely expressed in virtually all epithelium and is specifically found in tumors of epithelial origin. Matriptase has been implicated in carcinogenesis including ovarian, prostate and cervical cancers and is considered a drug target for developing cancer therapeutic agent.

SensoLyte® Rh110 Matriptase Assay Kit *Fluorimetric* - Cat# 72241
The SensoLyte® Rh110 Matriptase Assay Kit provides a convenient assay for screening of enzyme inhibitors and activators or for continuous assay of enzyme activity using a fluorogenic substrate. Upon matriptase cleavage, this substrate generates the Rh110 (Rhodamine 110) fluorophore which has a bright green fluorescence and can be detected at excitation/emission=496/520 nm. The longer wavelength spectra and higher extinction coefficient of Rh110 provide greater sensitivity and less interference from other reaction components. This assay can detect as low as 0.15 ng/mL active Matriptase.

Peptidyl-prolyl isomerase (PPIase) Pin1 catalyzes cis/trans isomerization of the phospho-Serine/Threonine-Proline peptide bonds. Pin1 consists of short N-terminal protein-protein interaction domain that allows enzyme to bind phosphoproteins and longer C-terminal izomerase domain. Pin1 has many biological substrates, plays critical role in cell-cycle regulation, and up-regulated in many human cancers. Recently, Pin1 was linked to the Alzheimer’s disease pathogenesis. Pin1 can bind phosphorylated Thr-212/231 residues of Tau protein and increase its dephosphorylation that may prevent formation of Tau neurofibrillary tangles.

SensoLyte® Green Pin1 Assay Kit *Fluorimetric* - Cat# 72240
The SensoLyte® Green Pin1 Assay Kit uses a fluorogenic substrate in the cis isoform. Pin1 changes substrate into trans conformation, which is then readily cleaved to generate fluorescent signal which can be monitored at Ex/Em=490/520 nm. Increase in fluorescence intensity is directly proportional to the Pin1 activity.

38 Amino Acid Preloaded Hydrazide Resins

Native Chemical Ligation (NCL) has been one of the most used bioconjugation methodologies for synthesis of macromolecules. NCL requires a peptide thioester and another peptide moiety containing a free cysteine at the N-terminus. However, peptide thioester synthesis is technically challenging due to its instability. The use of a peptide hydrazide is more recommended.

As a worldwide unique provider of Amino acid pre-loaded hydrazide resins, we offer a series of 38 ready-to-use hydrazide resins carrying one of the 19 natural-L or D isomer amino acids. These resins are perfectly suited for the production of activated peptide thioester via Solid Phase Peptide Synthesis (SPPS).
